How do syntax checks differ from mailbox checks?

Syntax checks only confirm that the address looks correct according to the rules defined in RFC 5322. Mailbox checks confirm whether the address actually exists by interacting with the receiving server. Syntax checks alone are shallow. Mailbox checks provide deeper insight but cannot always guarantee accuracy.
